Can anyone tell me if the Golden Tube Preamp Sep1 with 1 tube will make a difference in a system?

Yes it will.
You will notice the LPs sounding smoother and more musical in most cases.
You may also notice less ticks and pops. Solid state preamps tend to make more ticks and pops due to overload margin problems and sensitivities to RFI that tubes often don't have.

The reasons are pretty technical, but this is a very real phenomena!

Can anyone tell me if the Golden Tube Preamp Sep1 with 1 tube will make a difference in a system? 

I am also thinking about a pair of B&W 570 speakers or Bozak 300 to create a music only system. The speakers are 59.99 a pair, from the goodwill store and maybe an AKAI AM 2400 stereo amplifier. What do you think?  

I saw a Pioneer PL 200 and a Sanyo TP 1012 Turntable at my local goodwill store. I am thinking of picking up one of them.... 25.00 each.

What's the budget?

Different, yes. Not totally sure it will be much improved however. Which source do you use the most? If you want to play more records, start with improving the turntable to something like the Rega or Project or Pioneer PLX1000. Then upgrade your preamp to one with a good tube phono stage. The CD player will probably suffice for the moment. If you want to persue the 2 channel hifi route, I'd be inclined to say sell all your speakers and research getting a good single pair.
