In hooking up my speaker wire from amp to speaker is the red positive or negative? Thanks!

Red is only positive if hooked up as positive, and if left not hooked up to anything, it's nothing at all.
Out of phase speakers do matter...it's all about "suck in or suck out" and if out of phase they "suck in" and that sucks.
Hook up an electric bass cab backwards and see exactly what I mean. I worked with a bass player who did exactly that and the cab sounded pretty crappy until we re-wired it back to the proper polarity..."el sucko wrongo a mundo"...lower output, weird sound, and all of his fellow musicians pointing at him and laughing (not really as he was/is a very good bass player).
Try it. Any one speaker guitar or bass amp...hook it up backwards and it sounds bad. Period.
Stereo speakers if hooked up backwards will sound a little weird and the bass will suffer a lot...does it matter? Well...weird is weird, accurate is accurate.
If you hook up speakers backwards and play certain frequencies at high enought levels, it can trigger a rip in the "time continuim" of our current dimension of reality and end all life as we know it. I'm not saying that's necessarily a bad thing...
Let me know exactly when you do it so I can make sure I put on my tinfoil hat, as that provides the necessary insulation against such untoward activities. Make sure after you hook 'em up backwards you have time to duck behind the couch.
